Protein electrophoresis reagents and buffer related

 30%(W/V)acrylamide solution
* Component concentration 30% (W / V) Acrylamide
* Preparation of volume 100ml
* Preparation method 
1. The following reagents were weighed, placed in 250ml beaker.
Acrylamide       29g
BIS                  1g
2. Added to the beaker of about 80ml of deionized water, stir dissolved; final volume to 100ml, with a 0.45μm membrane filter impurity.
3. Placed in brown bottles at 4 ℃. (Note: a strong acrylamide neurotoxicity can be absorbed through the skin and are cumulative. Preparation should wear gloves.) 

 
40%(W/V)acrylamide solution
* Component concentration 40% (W / V) Acrylamide
* Preparation of volume 100ml
* Preparation Method 1. The following reagents were weighed, placed in 200ml beaker
Acrylamide       38g
BIS                  2g
2. Added to the beaker of about 80ml of deionized water, stir dissolved; final volume to 100ml, with a 0.45μm membrane filter impurity.
3. Placed in brown bottles at 4 ℃.
(Note: a strong acrylamide neurotoxicity can be absorbed through the skin and are cumulative. Preparation should wear gloves)

 
10%(W/V)ammonium persulfate
* Component concentration 10% (W / V) ammonium persulfate
* Preparation of volume 10ml
* Preparation Method 1. Weigh 1g of ammonium persulfate.
2. Adding 10ml of deionized water stirred solution, stored at 4 ℃. (Note: 10% ammonium persulfate solution at 4 ℃ to use about two weeks, after which the catalytic effect will be lost) 

 
5X Tris-Glycine Buffer
* Component concentration 0.125M Tris, 1.25M Glycine, 0.5% (W / V) SDS
* 1L * the amount of preparation method of preparation 1. The following reagents were weighed, placed in 1L beaker 
Tris             15.1g 
Glycine        94g 
SDS            5.0g
2. Add about 800ml of deionized water, stirring to dissolve. 3. Constant volume by adding deionized water to 1L, the room temperature 

 
2X SDS-PAGE Loading Buffer
* Component concentration
100mM          Tris-HCl (pH 6.8)
4% (W/V)      SDS
0.2% (W/V)   bromophenol blue
20% (VV)      glycerol
2% (W/V)     β-mercaptoethanol
* Preparation of volume 5ml
* Preparation method
1. The following reagents were weighed, placed in 15ml plastic centrifuge tube
1M Tris-HCl (pH 6.8)     0.5ml
SDS                              0.2g 
bromophenol blue       10mg
glycerol                       1ml
2. Deionized water solution volume to 5ml.
3. Small portion of repackaging, 0.5ml one, at room temperature. 4. Before using 25μl of β-mercaptoethanol added mix. 

 
5X SDS-PAGE Loading Buffer
* Component concentration
250mM          Tris-HCl (pH 6.8)
10% (W/V)   SDS
0.5% (W/V)  bromophenol blue
50% (V/V)    glycerol
5% (W/V)     β-mercaptoethanol
* * preparation method of preparation  Volume of 5ml
1. The following reagents were weighed, placed in 15ml plastic centrifuge tube
1M Tris-HCl (pH 6.8)         1.25ml
SDS                                  0.5g 
bromophenol blue           25mg
glycerol                            2.5ml
2. Deionized water solution volume to 5ml.
3. Small portion of repackaging, 0.5ml one, at room temperature.
4. Before using 25μl of β-mercaptoethanol added mix.
